Overview of the transport architecture
The major points that you need to understand from this diagram are the following:
Apart from local delivery to a database on the server, mailbox servers never transport mail. Instead, when a mailbox server has a message that it needs to send to a database hosted by another Exchange server or a recipient in another email system, the Microsoft Exchange Mail Submission service running on the mailbox server informs a hub transport server that it has a message to be fetched. The Submission service indicates the database, mailbox, and message identifier to point to where the message is waiting. The transfer of the message between mailbox and hub transport server is the responsibility of the Store driver.

The mailbox server performs the same processing for remote and local deliveries to ensure that all messaging compliance requirements are respected . In effect, outgoing mail goes into an Outbox folder in the Store where it is fetched by the Store driver, which moves the message to the hub transport server, where the message ends up in the submission queue . The transport service retrieves the messages from the submission queue, categorizes them, and then routes the mail by placing the messages onto the appropriate delivery queues for onward transmission .
Mailbox and hub transport servers within an Active Directory site communicate with each other using server-side remote procedure calls (RPCs). The need to accommodate Exchange RPCs requires the servers to be connected by local area network (LAN)-quality bandwidth because otherwise you run the risk of RPC timeouts and failure. If multiple hub transport servers are available in the site, the Mail Submission service selects one on a load-balancing basis. This provides a level of fault tolerance for the transport service. Unless the server is part of a DAG and another hub transport server is available within the site, if a server hosts both the mailbox and hub transport role, Exchange uses the local hub transport server to process messages. An exception is made when a server is part of a DAG because transferring messages off the mailbox server ensures that they are captured by the dumpster of the hub transport server and therefore can be recovered in the case of failure. The Active Directory Topology Service is responsible for providing the transport service with information about the current site topology by reference to an Active Directory domain controller/global catalog server.
